Overview

The mouse intermediate-affinity interleukin-2 receptor is a heterodimeric signaling complex composed of the interleukin-2 receptor subunit beta (CD122) and the common cytokine receptor gamma chain (CD132) (UniProt P16267, P34902). Unlike the high-affinity trimeric receptor that includes the alpha subunit (CD25), this intermediate-affinity form is primarily found on memory CD8+ T cells and natural killer (NK) cells in the murine immune system (PubMed PMID: 29434354). Activation of this receptor by interleukin-2 (IL-2) triggers the JAK1/JAK3 and STAT5 signaling pathways, which are essential for the proliferation and effector function of these cytotoxic immune cells (PubMed PMID: 10438930). In therapeutic research, this receptor is a critical target for developing biased IL-2 agonists designed to enhance anti-tumor immunity while minimizing the activation of regulatory T cells (Tregs), which preferentially express the high-affinity receptor (PubMed PMID: 30635424). Consequently, it serves as a focal point for studies involving IL-2 variants and immunotherapies aimed at treating cancer and chronic viral infections in mouse models and translational research.
