The H-2Kb MHC class I–Trp2 peptide complex is a molecular assembly consisting of the murine MHC class I molecule H-2Kb and a specific epitope derived from Tyrosinase-related protein 2 (Trp2), typically the 180-188 sequence (SVYDFFVWL) (Bloom et al., 1997). Trp2, also known as dopachrome tautomerase (DCT), is a melanosomal enzyme involved in melanin synthesis and is highly expressed in both normal melanocytes and melanoma cells, making it a significant tumor-associated antigen (Parkhurst et al., 1998). This complex plays a critical role in the immune system by presenting the Trp2 peptide on the surface of cells for recognition by the T-cell receptors (TCRs) of CD8+ cytotoxic T lymphocytes (Overwijk et al., 1999). In the context of oncology research, particularly using the B16 mouse melanoma model, this complex is a primary target for various immunotherapeutic strategies, including peptide-based vaccines, DNA vaccines, and adoptive cell therapies (Schreurs et al., 2000). By targeting this complex, therapies aim to stimulate a robust anti-tumor immune response capable of identifying and destroying melanoma cells. However, because Trp2 is also expressed in healthy melanocytes, targeting this complex can lead to autoimmune side effects such as vitiligo (Bowne et al., 1999).
