Overview

The MRE11-RAD50-NBS1 (MRN) complex is a highly conserved, multi-protein assembly essential for the detection and repair of DNA double-strand breaks (DSBs). The MRN complex plays central roles in sensing DSBs, processing DNA ends for repair, activating cell cycle checkpoint signaling via ATM/ATR kinases, and tethering and bridging broken DNA ends to facilitate accurate repair. It is comprised of the MRE11 nuclease, the RAD50 ATPase, and the NBS1 regulatory subunit. Mutations in MRN components are associated with severe genomic instability syndromes.
