Overview

The MRN complex-interacting protein (MRNIP) is a chromatin-binding protein that plays a critical role in the cellular response to DNA double-strand breaks and maintenance of genome stability[3][1][7]. MRNIP associates with the MRN complex (comprised of MRE11, RAD50, and NBS1) and promotes the loading and activity of the MRN complex at DNA damage sites, thereby enhancing ATM kinase activation and facilitating homologous recombination-mediated DNA repair[1][3][7]. MRNIP is capable of liquid-liquid phase separation, forming nuclear condensates that concentrate the MRN complex and damaged DNA to accelerate repair processes[1]. MRNIP regulates checkpoint signaling and is implicated in radioresistance in cancer models[1]. While MRNIP itself is not a current direct therapeutic target, its central role in genome maintenance and DNA repair positions it as a potential mediator of radiosensitivity or resistance in oncology[1][3][7].
