Overview

MSANTD2 antisense RNA 1 (MSANTD2-AS1), also known as BCLET (Bladder Cancer Low-Expressed Transcript) and RP11-677M14.7, is a nuclear-localized long non-coding RNA (lncRNA) that functions as the antisense transcript to the MSANTD2 gene. This lncRNA is significantly downregulated in human bladder cancer tissues and cells compared to normal controls. BCLET modulates the alternative splicing of the MSANTD2 gene, preferentially promoting the production of the MSANTD2-004 isoform. Overexpression of BCLET suppresses bladder cancer cell proliferation, invasive capability, and supports apoptosis, suggesting tumor suppressor-like activity. BCLET exerts its biological effects mainly through direct interaction with MSANTD2 mRNA, influencing splicing events crucial for tumor cell behavior. There is currently no evidence that BCLET serves as a direct therapeutic target or interacts with approved drugs, and it does not function as a receptor, enzyme, or classic druggable protein. Its potential role is primarily as a molecular mechanism and possible biomarker for bladder cancer risk and progression, rather than a classic therapeutic target[1][5].
