Overview

MT-ND4 pseudogene 3 (MTND4P3) is a nuclear-encoded pseudogene that shares sequence similarity with the mitochondrial gene MT-ND4. As a pseudogene, MTND4P3 does not encode a functional protein and is not implicated in classic biological pathways or diseases. Pseudogenes such as this one may arise from the transfer of mitochondrial genetic fragments to the nuclear genome, but mutations and premature stop codons render them nonfunctional—unable to produce an active protein product. MTND4P3 should not be confused with the functional mitochondrial gene MT-ND4, which encodes a core subunit of respiratory chain complex I. MTND4P3 is listed by the HUGO Gene Nomenclature Committee as a "Pseudogene" (HGNC:42191) and its pseudogene status is validated. Pseudogenes do not generally produce a protein, interact with drugs, or serve as classical disease markers or therapeutic targets. While detectable during genetic testing or analysis due to sequence similarity, their biological and clinical significance is negligible, except in rare cases where they might regulate gene expression via RNA interference (not shown for MTND4P3). MT-ND4 pseudogenes, including MTND4P3, may produce nonfunctional transcripts with multiple disabling mutations and do not contribute to mitochondrial complex I activity. This target is not considered a proper therapeutic target or receptor.
