Overview

MT-ND5 pseudogene 8 (MTND5P8) is a segment of nuclear DNA that originates from a partial duplication or transfer of the mitochondrial gene encoding NADH dehydrogenase 5 (MT-ND5) into the nuclear genome. This pseudogene, like other nuclear mitochondrial DNA segments (NUMTs), does not produce a functional protein. Instead, it closely resembles the sequence of its mitochondrial counterpart, which can lead to artifacts or errors in assays aimed at detecting mitochondrial DNA mutations. This has been noted in contexts such as cancer mutation analysis and mitochondrial disease studies, where distinguishing between NUMTs and true mitochondrial sequences is critical for accurate molecular diagnostics. MTND5P8 itself is not a therapeutic target, does not have known physiological or disease functions, and is not implicated in drug interactions or as a biomarker. MT-ND5 (not MTND5P8) is a functional gene encoding a component of mitochondrial complex I, involved in oxidative phosphorylation and associated with human disease when mutated. However, MTND5P8 is a pseudogene, not the functional mitochondrial gene. The presence of "pseudogene" in the name and its classification as a NUMT indicate it has no protein or enzymatic activity. Its relevance is primarily technical, regarding genetic analysis, rather than biological or therapeutic.
