MTSS I-BAR domain containing 2 (MTSS2), also called MTSS1L or ABBA, is a member of the I-BAR (Inverse Bin/Amphiphysin/Rvs) domain protein family, characterized by an N-terminal I-BAR domain that binds plasma membranes and induces negative membrane curvature, and a C-terminal actin-binding WH2 domain. MTSS2 is predominantly expressed in the central nervous system, especially in neurons, and is implicated in the formation and plasticity of synaptic structures such as dendritic spines. The protein participates in linking dynamic membrane shaping to actin cytoskeleton remodeling and has GTPase activator and small GTPase binding capacities. Human genetic variants in MTSS2 are associated with a syndromic form of intellectual disability, often including microcephaly and eye anomalies, suggesting its critical role in neurodevelopment and synaptic function. There are currently no drugs or established biomarkers directly targeting MTSS2, and it is not a recognized therapeutic target.
