Overview

The MUC1-derived peptide–HLA-A2 complex is a specific peptide-major histocompatibility complex (pMHC) formed when fragments of the Mucin 1 (MUC1) protein are presented by the HLA-A2 allele on the surface of cells. MUC1 is a transmembrane glycoprotein that is frequently overexpressed and aberrantly glycosylated in various epithelial malignancies, making it a prominent tumor-associated antigen. On dendritic cells, this complex serves as a critical immunological signal that primes and activates MUC1-specific CD8+ cytotoxic T lymphocytes (CTLs). These activated T cells then recognize and eliminate tumor cells that present the same pMHC epitope. Therapeutic strategies targeting this complex primarily involve dendritic cell-based vaccines and peptide-based immunotherapies designed to overcome immune tolerance and stimulate a robust anti-tumor response. Because the complex is restricted to the HLA-A2 allele, its clinical application is limited to patients carrying this specific genetic marker.
