MUC16xCD28 is a therapeutic target complex utilized in the development of costimulatory bispecific antibodies for the treatment of MUC16-positive malignancies, most notably ovarian cancer. Mucin 16 (MUC16), also known as CA-125, is a high-molecular-weight glycoprotein that is significantly overexpressed on the surface of ovarian cancer cells, making it an ideal tumor-associated antigen. CD28 is a critical costimulatory receptor on T cells that, when engaged, lowers the threshold for T-cell activation and promotes survival and cytokine production. By simultaneously binding to MUC16 on tumor cells and CD28 on T cells, this bispecific molecule provides the necessary 'Signal 2' to T cells that are already being primed by a 'Signal 1' (such as a CD3-engaging bispecific antibody). This synergistic approach is designed to overcome the immunosuppressive tumor microenvironment and drive a more robust and durable anti-tumor immune response compared to CD3-bispecific monotherapy alone.
MUC16xCD28 is a costimulatory bispecific antibody designed to bridge MUC16-expressing tumor cells with the CD28 receptor on T cells. It provides a potent secondary costimulatory signal (Signal 2) that enhances T-cell effector function and proliferation only when the T cell is simultaneously receiving a primary activation signal (Signal 1) through the T-cell receptor or a CD3-bispecific antibody (e.g., Ubamatamab). This dual-targeting approach aims to maximize anti-tumor activity while minimizing systemic cytokine release by restricting T-cell costimulation to the tumor microenvironment.
