Mucin-2 (MUC2) is a large gel-forming mucin and highly glycosylated protein produced by goblet cells of the intestinal tract. It is the primary constituent of the mucus layer that protects and lubricates the gastrointestinal epithelium, forming a physical and chemical barrier that separates the intestinal microbiota from the underlying tissues and immune system. The structure of MUC2 features a protein core of over 5000 amino acids, characterized by extensive tandem PTS (proline, threonine, serine-rich) repeats and a highly glycosylated central domain. The C-terminal end forms dimers through cystine-knot domains and additional disulfide bonds, contributing to stable mucus network formation. Besides its mechanical and protective role, MUC2 participates in regulating copper homeostasis and acts as a scaffold for antimicrobial molecules. Altered MUC2 function or expression is implicated in diseases such as inflammatory bowel disease and colorectal cancer, but it is not currently a direct drug target.
