Overview

Mucin-22 (MUC22) is a membrane-bound, high-molecular-weight glycoprotein belonging to the mucin family, expressed mainly on epithelial surfaces in the respiratory tract and other mucosal tissues[2]. It plays a crucial role in maintaining the mucosal barrier, modulating epithelial cell signaling, and suppressing cell proliferation and migration[1][2]. In non-small cell lung cancer (NSCLC), epigenetic modifications such as DNA methylation regulate MUC22 expression, with decreased expression acting as a poor prognostic marker in squamous cell carcinoma and increased expression marking poor prognosis in adenocarcinoma. While not currently targeted by any approved drugs, MUC22 and its epigenetic regulation are under investigation for their potential as diagnostic, prognostic, and therapeutic biomarkers in cancer—especially in precision medicine for NSCLC subtyping and risk assessment[1][2].

Mechanism of action

Epigenetic reactivation: Demethylating agents and histone deacetylase inhibitors can increase MUC22 expression by reducing promoter methylation and increasing histone acetylation[1].
