Overview

Mucin 6 (MUC6)-associated glycans, specifically those terminated with alpha-1,4-linked N-acetylglucosamine (alpha1,4-GlcNAc), are critical components of the human innate immune system within the gastric mucosa (UniProt Q13825). These unique glycans are primarily expressed in the mucous neck cells and pyloric glands of the stomach, where they are attached to the O-glycan side chains of the MUC6 protein. Their primary biological role is to act as a natural antibiotic against Helicobacter pylori by inhibiting the bacterial enzyme cholesterol alpha-glucosyltransferase, which is essential for the synthesis of the bacterial cell wall (Kawakubo et al., 2004, Science). In clinical contexts, a reduction in these glycans is associated with increased susceptibility to H. pylori infection and a higher risk of developing gastric adenocarcinoma (Nakayama et al., 2012, PNAS). Research into these glycans focuses on their potential as therapeutic agents or mimetics to treat antibiotic-resistant H. pylori infections and prevent gastric cancer progression. They also serve as important biomarkers for gastric health and the progression of pre-cancerous lesions, often detected using the HIK1083 antibody. The interaction between these glycans and H. pylori represents a unique example of host-derived carbohydrate-based antimicrobial defense that is highly specific to the gastric environment.
