Overview

Mucin 6 (MUC6)-associated glycans, specifically those terminated with alpha-1,4-linked N-acetylglucosamine (alpha1,4-GlcNAc), are specialized carbohydrate structures found in the gastric mucosa (Kawakubo et al., 2004, Science). These glycans are synthesized by the enzyme alpha-1,4-N-acetylglucosaminyltransferase (A4GNT) and are primarily localized to the mucous neck cells and pyloric glands (Nakayama et al., 2012, PNAS). Biologically, they serve as a natural antibiotic against Helicobacter pylori by inhibiting the synthesis of cholesteryl alpha-D-glucopyranoside, a vital component of the bacterium's cell wall (Kawakubo et al., 2004). In clinical contexts, the loss of these glycans is strongly associated with the development and progression of gastric adenocarcinoma, making them significant biomarkers for malignancy (Ishihara et al., 2017, Gastric Cancer). Therapeutic strategies involve using these glycans or their biosynthetic pathways to treat H. pylori infections or as targets for monoclonal antibodies in gastric cancer diagnostics and therapy (Hidaka et al., 2017, Cancer Science).
