Mucin disulfide bond hydrolysis refers to the enzymatic or chemical cleavage of disulfide (S–S) bonds within mucin proteins, primarily gel-forming secreted mucins like MUC2, MUC5AC, and MUC5B. These bonds are crucial for maintaining the polymeric structure and viscoelastic properties of mucus. Hydrolysis disrupts the mucin network, reducing mucus viscosity and elasticity, which can be therapeutically exploited to improve mucociliary clearance in diseases characterized by mucus hypersecretion or hyperviscosity, such as asthma, COPD, and cystic fibrosis. Agents like N-acetylcysteine promote this hydrolysis.
