The mucin glycoprotein layer is a structurally organized barrier comprised of heavily glycosylated mucin proteins that assemble into a viscoelastic, gel-like matrix at epithelial surfaces. This layer provides lubrication, regulates permeability to particles and pathogens, and acts as a first line of defense in innate immunity. Structural stabilization depends on non-covalent mucin-mucin interactions, extensive O-linked glycosylation, and environmental factors such as pH and ion concentration. Large mucins (e.g., MUC2, MUC5AC, MUC5B) form the structural backbone of the mucus gel, while membrane-associated mucins (e.g., MUC1) reinforce cell surface protection.
