Mucin polymers are large, heavily glycosylated proteins that form the structural framework of mucus gels. They are primarily composed of long polypeptide chains rich in proline, threonine, and serine (PTS domains), which undergo extensive O-glycosylation. These polymers assemble into complex networks via disulfide bonds and noncovalent interactions, creating viscoelastic gels that protect and lubricate mucosal surfaces. Key structural features include a bottlebrush-like architecture, high water-binding capacity, and resistance to proteolysis. The major gel-forming mucins in human airways are MUC5AC and MUC5B.
Variable; depends on the specific drug. Mucolytics break down mucus structure by disrupting disulfide bonds or other interactions. Expectorants increase mucus hydration and volume.
