Overview

Mucins bearing N-acetylglucosamine-alpha-1,4-galactose (alpha1,4-GlcNAc) glycotopes are specialized glycoproteins primarily found in the gastric mucosa, specifically on MUC6 mucins produced by mucous neck cells and gland cells (Kawakubo et al., 2004). These glycotopes are synthesized by the enzyme alpha-1,4-N-acetylglucosaminyltransferase (alpha4GnT) and serve as a critical component of the innate immune system in the stomach (Nakayama et al., 2012). Their primary biological role is to act as a natural antibiotic against Helicobacter pylori by inhibiting the bacterial enzyme cholesterol alpha-glucosyltransferase, thereby disrupting the synthesis of essential cell wall components (Kawakubo et al., 2004). In clinical contexts, the loss of these glycotopes is strongly associated with the progression of chronic gastritis to gastric adenocarcinoma, making them significant biomarkers for gastric cancer risk (Nakayama et al., 2012). Therapeutic strategies involve leveraging these glycotopes or their biosynthetic pathways to treat H. pylori infections and prevent gastric malignancy.
