Mucoadhesion and hydration of mucosal tissues refers to the property by which a material, typically a hydrophilic polymer, adheres to a mucous membrane and retains water. This process is crucial in the development of mucoadhesive drug delivery systems, allowing drugs or medical devices to remain at the administration site (such as the oral, nasal, vaginal, or gastrointestinal mucosa) for prolonged periods. Mucoadhesion involves a contact/initial wetting stage and a consolidation stage, with mechanisms including hydrogen bonding, Van der Waals forces, polymer-mucin interpenetration (diffusion theory), mechanical entanglement, and occasionally electrostatic interactions. Hydration enables the polymer to swell, maximizing contact with the mucosal surface, increasing viscosity, and facilitating physical entanglement with mucins. These properties aid in controlled drug release, enhanced bioavailability, and improved therapeutic efficacy. However, mucoadhesion is a property of materials and not a distinct therapeutic target; it is achieved through judicious material and formulation design rather than receptor-ligand molecular targeting.
