Mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1) is a paracaspase enzyme that plays a central role in immune cell signaling, particularly in the activation of lymphocytes. It is essential for both the immune response and the development of regulatory T (Treg) cells that help maintain immune homeostasis. MALT1 acts as both a scaffold and a protease within the CARMA1-BCL10-MALT1 (CBM) signalosome complex, which is crucial for triggering NF-κB signaling pathways. Exaggerated activity of MALT1 has been linked to lymphoid malignancies, autoimmunity, and inflammation. Small-molecule inhibitors targeting its proteolytic activity are being explored as potential therapies.
