Overview

The mucosal/epithelial surface interface is a multi-layered biological boundary that separates the body's internal tissues from the external environment or luminal spaces, such as the gastrointestinal, respiratory, and urogenital tracts. It is composed of a specialized epithelial cell layer, interconnected by tight junctions, and overlaid by a protective mucus gel containing mucins, antimicrobial peptides, and secretory antibodies (NIH, 2023). This interface plays a dual role: it acts as a selective filter for nutrient and gas exchange while serving as a robust physical and immunological barrier against pathogens, toxins, and environmental insults (Nature Reviews Gastroenterology & Hepatology, 2020). Dysfunction of this interface, often characterized by increased permeability or leakiness, is central to the pathogenesis of numerous conditions, including inflammatory bowel disease, asthma, and various infections (PubMed, 2021). Although it is a physiological site rather than a single protein target, therapeutic interventions often focus on reinforcing its structural integrity or modulating the local immune environment to restore homeostasis (StatPearls, 2023).
