Mucosal immune system enhancement refers to the physiological process of strengthening immune defenses at mucosal surfaces, such as the gastrointestinal, respiratory, and urogenital tracts, to prevent pathogen entry and maintain homeostasis (Mowat & Agace, 2014, Science). This approach is particularly relevant for infectious diseases, where localized immunity can provide a first line of defense that systemic immunity might miss. However, the challenge in drug development lies in achieving potent activation without triggering chronic inflammation, autoimmunity, or the induction of oral tolerance, which can negate the intended protective effect. Consequently, while 'mucosal immune system enhancement' describes a therapeutic goal, drug discovery efforts focus on specific molecular triggers within the mucosal environment to achieve this state.
