Overview

Multi-PDZ domain protein 1 (MPDZ), also known as MUPP1, is a large scaffolding protein containing 13 PDZ domains that facilitate diverse protein-protein interactions. It is primarily localized at tight junctions in epithelial and endothelial cells, where it organizes signaling complexes and maintains barrier integrity by interacting with proteins like claudins and junctional adhesion molecules (UniProt, 2024). Beyond its structural role, MPDZ regulates the trafficking and signaling of various G protein-coupled receptors; for instance, its 10th PDZ domain binds to the 5-HT2C receptor, influencing synaptic plasticity and neurotransmission (PubMed, 2010). Mutations in the MPDZ gene are a known cause of autosomal recessive congenital hydrocephalus, a condition characterized by the accumulation of cerebrospinal fluid in the brain (NCBI Gene, 2023). In the context of oncology, specific PDZ domains of MPDZ (such as 7, 8, and 11) are targeted for degradation by high-risk human papillomavirus (HPV) E6 oncoproteins, contributing to cellular transformation and viral pathogenesis (PubMed, 2015). While no FDA-approved drugs currently target MPDZ, its PDZ domains are being explored as therapeutic targets using peptide-based inhibitors to disrupt specific pathological protein interactions.
