A multi-target network is a systems pharmacology concept referring to the network of molecular targets (such as proteins, receptors, enzymes) within a biological system that are simultaneously or sequentially modulated by a drug or combination of drugs. This framework underlies the development of multi-target drugs or network therapeutics, which aim to modulate several nodes (targets) in a biological network to treat complex diseases with multifactorial etiologies, such as cancer, neurodegenerative diseases, and metabolic syndromes. Whereas classical pharmacology focuses on a single molecular target, network pharmacology and multi-target approaches leverage computational, bioinformatics, and systems biology methods to predict, model, and validate drug actions across the entire network of disease-associated molecules. Multi-target drugs often exhibit lower affinity at individual targets but may result in higher efficacy and lower side effects by modulating several key pathways simultaneously. Such approaches can overcome drug resistance and redundancy mechanisms seen in complex diseases, but they also present challenges in drug design, safety prediction, and regulatory assessment due to the complexity of network interactions[1][5][7][8].
