MCIDAS (Multiciliate differentiation and DNA synthesis associated cell cycle protein, also known as Multicilin) is a small, coiled-coil, nuclear protein that acts as a transcriptional coregulator essential for the differentiation of multiciliated epithelial cells. It functions by forming a complex with transcription factors E2F4/E2F5 and their cofactor Dp1 (the "EDM complex"), activating genes required for massive centriole biogenesis and the deuterosome pathway, which are critical for multiciliogenesis. MCIDAS is upstream of central regulators of ciliary motility such as CCNO and FOXJ1, and its loss-of-function mutations cause congenital disorders due to defective mucociliary clearance, including RGMC. MCIDAS does not directly bind DNA but coactivates transcription through protein-protein interactions with other transcription factors.
