Overview

Multifunctional ROCO family signaling regulator 1 (MFHAS1) is a **probable GTP-binding protein** that plays regulatory roles in innate immunity, particularly by modulating **Toll-like receptor (TLR2 and TLR4) signaling pathways**, impacting inflammatory responses[1][11]. MFHAS1 can both inhibit and activate immune pathways: it negatively regulates TLR4-driven AP-1 activation by controlling phosphatase complex localization and positively modulates TLR2-dependent pro-inflammatory signaling, especially via p38/JNK MAP kinase activation[1][8][11]. It is also implicated in erythrocyte development and may play a role in inflammation induced by high glucose through AKT signaling[1]. MFHAS1 harbors structural features such as an ATP/GTP-binding site, leucine zippers, and leucine-rich repeats[1]. It has been described as a potential oncogene, especially in tumors with 8p amplification such as certain malignant fibrous histiocytomas, but there is no current evidence for therapeutic targeting with specific drugs or its use as a clinical biomarker[1][6][11].
