Overview

Multiple C2 and transmembrane domain-containing protein 1 (MCTP1) is a member of a conserved protein family that contains three high-affinity calcium-binding C2 domains and two transmembrane regions, anchoring it permanently in intracellular membranes, including the endoplasmic reticulum[1][2][4][5]. MCTP1 is highly expressed in the central nervous system, particularly in neuronal cell bodies and pre-synaptic axon terminals, where it localizes to secretory vesicles and endosomes[1][2]. Its unique C2 domains function as calcium sensors, influencing synaptic vesicle retrieval and homeostatic synaptic plasticity, and regulating endocytic recycling processes at neuronal synapses[1][2][4]. MCTP1 also plays a role in lipid droplet biogenesis by remodeling ER membrane curvature and facilitating contacts between organelles[5][7]. Genetic variants in MCTP1 have been associated with neuropsychiatric disorders such as bipolar disorder[2]. No established small-molecule or biologic drugs directly target this protein, nor is it used clinically as a biomarker.
