Overview

Multiple epidermal growth factor-like domains protein 11 (MEGF11) is a transmembrane protein characterized by several epidermal growth factor (EGF)-like domains. It is involved in homotypic cell-cell adhesion and plays a critical role in retina layer formation and the spacing (mosaic) of specific neuron subtypes through repulsion mechanisms. MEGF11 is implicated in human diseases such as congenital myopathy and anemia and is proposed as a novel therapeutic target in oncology, particularly for triple-negative breast cancer, where its overexpression is linked to tumor recurrence, modulation of cytokine and chemokine cascades, and poor prognosis. MEGF11 may also have potential (predicted) scavenger receptor activity and participates in pathways involving AKT, mTOR, NF-κB, and CREB/AP-1, though it does not co-localize with EGFR or Gs protein in tumor cells.
