Multiple epidermal growth factor-like domains protein 6 (MEGF6) is a large, secreted protein characterized by multiple epidermal growth factor (EGF)-like domains, some of which have calcium-binding capacity[1][3]. MEGF6 is predicted to enable calcium ion binding and scavenger receptor activity, and it is thought to be involved in vesicle-mediated transport in the extracellular region[1]. In cancer biology, MEGF6 promotes the epithelial-to-mesenchymal transition (EMT) via activation of the TGFβ/SMAD signaling pathway, facilitating metastasis and tumor progression, especially in colorectal cancer[1][3]. Rare variants in MEGF6 that disrupt its EGF-like calcium-binding domains have been linked to increased risk of osteoporosis, likely through impairment of cartilage and bone formation[1][3]. Functional studies in zebrafish support a crucial role for MEGF6 in skeletal development, mineralization, and possibly calcium homeostasis[3]. MEGF6 is not currently established as a therapeutic target or known receptor, but rather as an extracellular signaling and structural protein affecting disease processes in bone and cancer[1][3].
