Overview

Multiple Helicobacter pylori proteins represent the collective therapeutic targets within the Helicobacter pylori bacterium, a Gram-negative pathogen that colonizes the human gastric mucosa. Key targets within this group include urease, which neutralizes gastric acid to allow survival; penicillin-binding proteins (PBPs) involved in cell wall synthesis; the 30S and 50S ribosomal subunits responsible for protein translation; and DNA gyrase, which is essential for DNA replication. These proteins are the primary targets for standard triple and quadruple antibiotic therapies used to eradicate the infection. Helicobacter pylori is a major causative agent of chronic gastritis, peptic ulcers, and is classified as a Group 1 carcinogen due to its role in gastric cancer and MALT lymphoma. Effective targeting of these proteins is increasingly challenged by the global rise of multi-drug resistant strains, necessitating the development of novel inhibitors and vaccine candidates targeting specific virulence factors like CagA and VacA.
