Overview

The term Multiple immune and hematopoietic protein targets refers to a collective group of cell surface antigens primarily expressed on T-lymphocytes, but also found on B-cells, natural killer cells, and other hematopoietic lineages. This target set includes various Cluster of Differentiation (CD) molecules such as CD2, CD3, CD4, CD8, CD11a, CD18, CD25, CD44, and CD45, as well as HLA class I and II molecules. These proteins are essential for immune recognition, T-cell activation, and cellular adhesion during the immune response. In a therapeutic context, these targets are collectively addressed by polyclonal antibody preparations like Antithymocyte Globulin (ATG). By binding to this diverse array of receptors and markers, these drugs facilitate the profound depletion of reactive T-cells and the modulation of immune function, making them critical in the prevention and treatment of acute organ transplant rejection, graft-versus-host disease (GVHD), and severe aplastic anemia.

Mechanism of action

Polyclonal antibodies bind to a broad spectrum of surface proteins on T-lymphocytes and other hematopoietic cells, inducing rapid cell depletion through complement-dependent cytotoxicity (CDC), antibody-dependent cellular cytotoxicity (ADCC), and apoptosis, while also modulating immune signaling and cell trafficking.
