Overview

Multiple low-molecular-weight luminal solutes refer to a collective group of osmotically active substances within the gastrointestinal tract that facilitate water retention through physical-chemical properties. This 'target' is primarily associated with the pharmacology of osmotic laxatives, such as lactulose, which is metabolized by colonic bacteria into organic acids like lactic and acetic acid (DrugBank DB00581). These solutes increase the osmotic pressure in the colon, drawing water into the lumen to increase stool volume and soften its consistency, thereby promoting defecation (StatPearls, Osmotic Laxatives, 2023). In addition to treating constipation, the accumulation of these solutes and the subsequent acidification of the luminal environment are used to manage hepatic encephalopathy by trapping ammonia as non-absorbable ammonium ions (PubMed, PMID: 28260266). Because this target represents a collection of molecules acting via an osmotic gradient rather than a specific protein or receptor, it is classified as a non-specific therapeutic target.

Mechanism of action

These solutes increase the osmotic pressure within the intestinal lumen, creating a gradient that draws water into the bowel to increase stool volume, soften consistency, and stimulate peristalsis.
