Overview

Multiple myeloma tumor-associated protein 2 (C1orf35) is a human protein encoded by the C1orf35 gene, also named chromosome 1 open reading frame 35. Identified as a candidate oncogene from multiple myeloma cell lines, C1orf35 promotes tumor cell proliferation, accelerates cell cycle progression from G1 to S phase, and activates c-MYC transcription by binding the i-motif in the NHE III1 region of the c-MYC promoter. Its gene/protein is notably upregulated in multiple myeloma and shows positive association with disease progression, highlighting its role in oncogenesis, particularly in c-MYC–driven malignancies[1][2][4]. However, its broader molecular functions, classification, and therapeutic relevance are incompletely characterized. There are no established drugs directly targeting this protein, and it is not currently a validated therapeutic target.
