Overview

MPDZ (Multiple PDZ domain crumbs cell polarity complex component) is a large scaffold protein composed of up to thirteen PDZ domains, facilitating the assembly of diverse protein complexes at cellular junctions. It regulates cell polarity, cytoskeletal organization, signal transduction, and receptor localization—particularly clustering the 5-HT2C serotonin receptor at the cell membrane surface. MPDZ plays crucial roles in maintaining ependymal cell integrity in the brain, regulates sperm motility through transcriptional control of the CatSper cation channel, and influences cell migration in contexts such as cancer and neural signaling. Mutations or loss of MPDZ are associated with congenital hydrocephalus, certain eye disorders, and altered responses to behavioral stimuli such as ethanol. While MPDZ itself is generally not a direct pharmacological target, its interactors and modulatory activities at the cell surface and in signaling pathways are of potential therapeutic interest.
