Overview

Multivesicular body subunit 12A (MVB12A) is a protein component of the ESCRT-I (Endosomal Sorting Complex Required for Transport-I) complex that plays a central role in endosomal sorting and the biogenesis of multivesicular bodies[1][2]. MVB12A enables lipid and ubiquitin binding, and is involved in the regulation of epidermal growth factor receptor (EGFR) signaling by mediating cargo sorting into MVBs prior to lysosomal degradation[1][2]. It is essential for efficient vesicular trafficking, interacts with other ESCRT-I components (such as TSG101, VPS28, and VPS37), and enhances the sorting of endocytic ubiquitinated cargos [1][2]. MVB12A has been associated with diseases involving dysregulation of endosomal trafficking, such as atypical hemolytic-uremic syndrome and viral budding during HIV infection. While not a direct therapeutic target, MVB12A is critical in pathways relevant to cellular signaling and viral pathogenesis[1][2].
