Overview

Multivesicular body subunit 12B (MVB12B) is a protein that acts as a component of the ESCRT-I complex, a heterotetramer involved in regulating intracellular vesicular trafficking, particularly the sorting of ubiquitinated proteins from the plasma membrane to endosomal vesicles[2][8]. The ESCRT-I complex is essential for protein transport at endosomes, cytokinesis, and viral budding, notably in the HIV life cycle[2][8]. MVB12B specifically binds acidic phospholipids via its MABP domain, localizes to subcellular puncta and the plasma membrane, and interacts with core ESCRT-I subunits (TSG101, VPS28, VPS37 family) through conserved binding motifs[1][5][8]. While MVB12B is important for membrane trafficking and viral egress, there are currently no drugs, biomarker utilities, or established therapeutic targeting approaches associated with this protein[2][8].
