Overview

Mumps orthorubulavirus (MuV) is an enveloped, single-stranded, negative-sense RNA virus belonging to the Paramyxoviridae family and is the causative agent of mumps (StatPearls, 2023). The virus primarily targets the respiratory tract and salivary glands, leading to characteristic parotitis, but can also disseminate to the central nervous system and reproductive organs (CDC, 2021). Its genome encodes several structural proteins, including the hemagglutinin-neuraminidase (HN) and fusion (F) proteins, which are critical for host cell attachment and membrane fusion (PubMed, PMC4147624). The host immune system responds to MuV through innate interferon signaling and the production of neutralizing antibodies that target these surface glycoproteins (UniProt, 2024). While the MMR vaccine has significantly reduced the global incidence of mumps, the virus remains a therapeutic target for vaccine development and potential antiviral research due to its ability to cause outbreaks in vaccinated populations and its role in serious complications like orchitis and meningitis (NIH, 2022).
