Murine double minute 2 (MDM2), also known as E3 ubiquitin-protein ligase Mdm2, is a key regulatory protein that primarily functions as an E3 ubiquitin ligase targeting the tumor suppressor p53 for proteasomal degradation. MDM2 inhibits p53's transcriptional activity, promoting its ubiquitination. Overexpression or amplification of MDM2 has been observed in various cancers, disabling the tumor-suppressive functions of p53. Beyond p53 regulation, MDM2 can also target other proteins contributing to cell proliferation and survival. Due to its central role in cancer progression, antagonists targeting the interaction between MDM2 and p53 are under active investigation as anti-cancer therapies.
