The muscarinic acetylcholine receptor M5 (M5 mAChR) is a G protein-coupled receptor that mediates the effects of acetylcholine in the central and peripheral nervous systems. Primarily expressed in the brain, particularly on dopamine neurons, it modulates dopamine release and plays a role in behavioral flexibility and reward pathways. It is a potential therapeutic target for drug addiction and may be involved in Parkinson's disease pathophysiology.
Agonist binding leads to Gq/11 activation, PLCβ activation, IP₃/DAG production, and increased intracellular calcium. Inverse agonists like tiotropium reduce basal receptor activity.
