Overview

Musculin (MSC) is a basic helix-loop-helix transcriptional repressor capable of binding E-box elements as a homodimer or as a heterodimer with E2A proteins such as TCF3/E47[1][6]. It primarily functions to inhibit the transcriptional activity of E47, thereby acting as a negative regulator of antigen-dependent B-cell differentiation and is a downstream target of the B-cell receptor signal transduction pathway[1]. It also modulates human T-helper 17 cell responses to interleukin 2, influencing STAT5B activity in immune regulation[5]. Musculin is associated with diseases such as rhabdomyosarcoma and severe congenital neutropenia, X-linked[1]. Its paralog is TCF21, and it may play context-specific roles in cellular development and immune cell differentiation[2][4][5].
