Overview

MV4-11 is a human acute myeloid leukemia (AML) cell line derived from the blast cells of a 10-year-old male patient with biphenotypic B-myelomonocytic leukemia (ATCC, CRL-9591). It is a critical tool in hematology research due to its specific genetic profile, most notably the presence of the t(4;11)(q21;q23) translocation which creates the MLL-AF4 (KMT2A-AFF1) fusion protein (DSMZ, ACC 102). Additionally, MV4-11 cells harbor a homozygous internal tandem duplication (ITD) mutation in the FLT3 gene, making them highly sensitive to FLT3 kinase inhibitors (PubMed, PMID: 11060057). Because of these features, the cell line is frequently employed in preclinical studies to evaluate the efficacy of novel small-molecule inhibitors and to understand the molecular mechanisms of leukemogenesis (PubMed, PMID: 15143071). It is not a therapeutic target itself but rather a biological model used to study targets like FLT3 and MLL. The cell line is often used in both in vitro assays and in vivo xenograft models to assess drug potency and pharmacodynamics. Its consistent growth characteristics and well-defined genetic background make it a gold standard for AML research.
