Overview

Myb-binding protein 1A (MYBBP1A) is a nucleolar protein encoded by the MYBBP1A gene on human chromosome 17. It functions as a transcriptional co-regulator, both repressing and activating gene expression by interacting with DNA-binding proteins. It is involved in regulation of rRNA synthesis, cellular response to stress, modulation of the circadian clock (notably repression of the PER2 gene), and supports tumor suppressor functions by enhancing the activity of p53. MYBBP1A preferentially binds to specific chromatin marks (e.g., dimethylated histone H3 Lys-9) and contributes to epigenetic regulation. Although MYBBP1A is biologically significant and linked to important cellular processes and diseases such as cancer, it is not a current therapeutic target and no drugs are known to specifically act on this protein

Mechanism of action

None established. Since no drugs specifically target MYBBP1A, there are no defined mechanisms of action for this protein as a drug target

03

Biological functions

Regulation of transcription (activation and repression, including via interaction with sequence-specific DNA-binding proteins)Transcription corepressor activityRegulation of ribosomal DNA synthesis (rRNA biogenesis)Response to nucleolar stressTumor suppression (enhances p53 tetramerization and activation)Regulation of circadian clock gene expression (inhibits PER2 transcription via chromatin modification)Positive regulation of anoikis (programmed cell death due to loss of anchorage)Cellular response to glucose starvation
04

Disease associations

Cancer (influences tumor suppression through p53 pathway and may regulate cellular processes relevant to malignancy)Other (possible roles in metabolic stress and cell death, but main disease implication is in oncogenesis and tumor biology)
