Myb domain protein 4 (MYB4) is a transcription factor belonging to the R2R3-MYB subfamily, primarily characterized in the model plant Arabidopsis thaliana. It functions as a transcriptional repressor that modulates the phenylpropanoid metabolic pathway by inhibiting the expression of genes such as cinnamate 4-hydroxylase (C4H). This regulation controls the production of secondary metabolites like sinapate esters and flavonoids, which are essential for environmental adaptation, specifically providing protection against UV-B radiation, drought, and heavy metal stress. MYB4 is not a human protein and is not a recognized therapeutic target in human medicine. While vertebrate MYB family members (such as c-MYB/MYB, MYBL1/A-MYB, and MYBL2/B-MYB) are significant therapeutic targets in human cancers like leukemia and adenoid cystic carcinoma, they are not canonically referred to as 'MYB4'. The designation 'MYB4' in research is either specific to plant biology or used as an index for specific MYB-derived peptides in immunological studies. Experimental research has explored using plant-derived nanovesicles to deliver microRNAs that target MYB4-like pathways to influence processes like melanogenesis in mammalian cells, but these remain early-stage cross-kingdom studies rather than established therapeutic applications.
