MYB (myeloblastosis) is a proto-oncogene encoding a transcription factor essential for hematopoiesis, cell proliferation, and differentiation. It regulates gene expression by binding to specific DNA sequences. Aberrant expression or rearrangements of MYB are implicated in various cancers, particularly leukemias and lymphomas. It contains three helix-turn-helix (HTH) DNA-binding domains, a central transcriptional activation domain, and a C-terminal regulatory/repression domain. Its unique functional plasticity allows minor mutations to dramatically alter its regulated genes, underlying both normal physiological roles and oncogenic potential.
