Target intelligence / Profile preview

MYC proto-oncogene protein (MYC)

Target
MYC
Molecular classification
Transcription factor, bHLH-LZ transcription factor
01

Overview

MYC is a basic helix-loop-helix leucine zipper (bHLH-LZ) transcription factor that regulates the expression of thousands of genes involved in cell cycle progression, cell growth, apoptosis, differentiation, stem cell self-renewal, and metabolism. It is frequently deregulated in human cancers, leading to uncontrolled cellular proliferation and tumor aggressiveness. MYC also modulates the immune response within the tumor microenvironment, regulating immune checkpoint molecules and influencing both innate and adaptive immunity.

c-MycMYC
02

Mechanism of action

MYC activates gene expression by binding enhancer box sequences (E-boxes) and recruiting histone acetyltransferases, and it can repress gene expression by interacting with other transcription factors.

Safety considerations

MYC is a difficult therapeutic target due to its ubiquitous expression and essential role in normal cellular processes.Inhibition of MYC can lead to significant off-target effects.MYC overexpression can trigger apoptosis in non-transformed cells, creating a complex therapeutic window.

Biomarkers

MYC expression levels can be a biomarker for cancer diagnosis and prognosis
