These components include secreted proteins, lipids, cell wall molecules, and live or inactivated bacteria, each capable of engaging different immune pathways. Key molecular components include secreted proteins (MPB70, MPB83), lipids (mycosid B), and cell wall components like phosphatidylinositol mannosides and the 19-kDa lipoprotein. These components engage pattern recognition receptors like TLRs (especially TLR2) and C-type lectin receptors on APCs, leading to the activation of cytokine pathways involving Th1 cytokines (IFN-γ), Th2 cytokines (IL-4), iNOS, and IL-22. Cellular targets include dendritic cells, macrophages, NK cells, and neutrophils. Functional outcomes include enhanced IFN‑γ production, increased iNOS/IL‑22 levels, neutrophil priming, induction of Th1-biased response, and boosted IgG/IgM antibody production. Stimulation with purified protein derivatives is used in tuberculin skin tests, and BCG therapy exploits these immunostimulatory properties.
