Overview

Mycobacterium protein tyrosine phosphatase B (mPTPB), also known as PtpB, is a secreted enzyme produced by *Mycobacterium tuberculosis* (Mtb). It is an essential virulence factor that modulates host signaling pathways to promote bacterial survival inside macrophages. It exhibits triple-specificity phosphatase activity and dephosphorylates tyrosine, serine, and threonine residues. mPTPB subverts innate immune responses, blocks ERK1/2 and p38 MAP kinase-mediated IL-6 production, and promotes host cell survival via Akt pathway activation. It is a validated drug target with inhibitors showing promise as anti-TB agents.
