Myelin-associated glycoprotein (MAG), Reticulon 4 (Nogo protein), and Oligodendrocyte myelin glycoprotein (OMGP) are three distinct, membrane-associated inhibitors of axonal regeneration in the central nervous system. MAG, Nogo (Reticulon 4), and Oligodendrocyte myelin glycoprotein (OMGP) primarily inhibit axonal regeneration in the central nervous system. They largely converge on shared receptor systems, mainly the Nogo receptor (NgR1), and activate downstream inhibitory signaling pathways, notably the Rho kinase (ROCK) pathway. MAG also binds to neuronal gangliosides (GD1a, GT1b), Nogo receptor 2 (NgR2), β1-integrin, and PirB.
