Myelin basic protein peptide antigens are linear peptide fragments derived from the myelin basic protein (MBP), a key structural component of the myelin sheath in the central nervous system. These peptides are important immunodominant antigens, recognized by both T cells and B cells, and are central to the pathogenesis of autoimmune demyelinating diseases like multiple sclerosis (MS). Specific MBP peptides—including regions corresponding to amino acids 81–106, 83–101, 87–106, 89–99, among others—are recognized by autoreactive T cells in MS patients due to their presentation by MHC class II molecules such as HLA-DR2 and class I molecules such as HLA-A2 and HLA-B27. The immunodominant MBP peptides contribute to tolerance escape and initiation of CNS-targeted autoimmune responses. These antigens serve as models in animal and in vitro systems for understanding MS pathogenesis and for the development and testing of immunomodulatory therapies.
