Overview

The myelin basic protein (MBP)-specific T cell response refers to the immune activity of T lymphocytes that recognize and react to MBP, a major component of the myelin sheath surrounding nerve fibers in the central nervous system. This response is highly relevant in autoimmune diseases such as multiple sclerosis (MS), where autoreactive T cells target MBP, leading to demyelination and neurological dysfunction. Both CD4+ helper and CD8+ cytotoxic T cells can be specific for MBP. Autoreactive T cells recognize specific peptide fragments derived from MBP presented on MHC molecules by antigen-presenting cells. Activation can occur via molecular mimicry: microbial peptides with sequence similarity to MBP may trigger cross-reactive responses that break tolerance and initiate autoimmunity. Central and peripheral tolerance mechanisms exist but are incomplete; self-reactive lymphocytes specific for MBP persist even in healthy individuals.
